Der Krieg in Europa ist vorbei, doch die letzten Worte eines sterbenden Mannes führen Indy auf ein neues Abenteuer: die Suche nach einem der größten Schätze Alexanders des Großen! Verfolgt von einem gefährlichen Einäugigen folgt Indy der Spur eines Diamanten, die ihn von London über Alexandria in die Südsee führt, wo er auf eine mörderische Bande chinesischer Piraten trifft. Nach einer spektakulären Seeschlacht wird Indy von den Piraten auf einer einsamen Insel ausgesetzt, wo er wilden Kopfjägern in die Hände fällt, doch bevor ihn diese in einen Schrumpfkopf verwandeln können, kommt ihm der Anthropologe Bronislaw Malinowski zur Hilfe, und Indy trifft eine Entscheidung, die sein Leben verändert.
